Technical Abstract: For detecting subgroup J avian leukosis virus (ALV-J) specifically, the N terminus of envelope gene, containing vr1, hr1, hr2, of a Taiwanese strain 2921/00 was cloned into pRSET vector (pRSET/2921/gp85N plasmid) and expressed in BL21 E. coli. The expressed protein, pRSET/2921/gp85N, reacted with anti-ALV-J antiserum but not with subgroup A or B of avian leukosis virus. This vector was injected into mice several times for monoclonal antibody preparation. Two monoclonal antibodies were obtained, MAb14 AND MAb22. Both monoclonal antibodies reacted with pRSET/2921/gp85N protein but only the latter was reacted with infectious virion of ALV-J by Western blotting. In antigen capture ELISA, MAB22 served as a capture antibody better than as a coating antibody.
