Technical Abstract: Here we provide detailed protocols for preparing flowers for microscopic observation of pollen and for collecting pollen in bulk for germination or for RNA or protein isolation. We outline crossing schemes that will help confirm or refute hypothesized roles for the gene of interest, and we outline phenotypic characterizations that are useful in determining whether a gene product is important during one or more phases of pollen development or germination.
