Objective:
1) Define and enhance the immune response to acute and chronic Salmonella infection in swine; 2) Determine the effect(s) of transportation stress on the neuroendocrine and immune systems of swine; 3) Identify virulence factor(s) and/or mechanisms important in the pathogenesis of S. choleraesuis and S. typhimurium in swine. Previously was 3625-42000-010-00D (05/01).

Approach:
Immune parameters associated with salmonellosis will be measured by bioassay, ELISA, chemiluminescence, and flow cytometry. The interaction between stress, porcine immunity, and Salmonella spp. shedding, will be evaluated using a porcine stress model. Neuroendocrine responses will be measured in collaboration with the Livestock Behavior Research Unit at Purdue University. A bactericidal assay using flow cytometry will be developed and compared with genetic polymorphisms. Porcine mRNA expression will be analyzed using cDNA microarray chip technology. Production of biologically active porcine sTNFR-1 will be optimized. Virulence factors and/or mechanisms underlying pathogenesis of Salmonella spp. (including S. typhimurium DT104) in the gut will be identified using electron microscopy and histological techniques.
