Research Project: Estrogen Progesterone Modulation of Nitric Oxide Production in Endotoxemic Calves

Objective:
(1)Determine the mechanism of action by which the administration of estrogen and progesterone to calves in the form of a sustainable release implant limits the extent of tissue damage associated with the production of nitrogen and oxygen-derived free radicals. (2) Assess the utility of this steroid treatment to modify host response to infection stress in a beneficial, cost-effective manner.

Approach:
Tissues and various biological fluids will be analyzed for the expression and presence of different forms of nitric oxide synthase using the differential enzyme assay developed and validated in our laboratory. In addition immunohistochemical procedures will be employed to assess site specificity of oxidative nitration reaction in different cell as well as the cellular ditribution of nitric oxide synthase isoforms. The level of plasma tumor necrosis factor-alpha will be quantified by radioimmunoassay.
