Objective:
Characterize the genes for soluble acid invertase, sucrose synthase, and other genes of sucrose metabolism that correlate to sucrose concentration in sugarcane. Test the hypotheses that differences in expression of soluble acid invertase and sucrose synthase-1 between Saccharum species contribute to difference in sucrose accumulation in those species, and that differences in the promoter regions of these genes are responsible for the differences in expression.
Approach:
Isolate the entire gene for SAI and SuSY1 from a high-sucrose S. officinarum and a low-sucrose S. spontaneum. Determine allelic differences within and between genotypes. Screen progeny from a cross of the two genotypes for the different alleles. Identify and characterize other SAI and SuSy genes. Investigate regulation of SAI genes by in callus culture. Identify gene expression related to natural ripening in sugarcane. Develop a cDNA library enriched in ripening-specific genes. Identify ripening specific genes. Verify changes in expression of candidate genes during ripening.
