Research Project:
PRODUCTION OF GERMPLASM AND MOLECULAR RESOURCES FOR WHEAT IMPROVEMENT
Location:
Crop Improvement/utilization Research
Project Number: 5325-21430-006-00
Project Type:
Appropriated
Start Date: Oct 01, 1999
End Date: Sep 30, 2004
Objective:
To produce wheat with improved agronomic and end-use traits, including better food processing quality. To increase the Fusarium head blight resistance of wheat by developing and applying biotechnology resources and protocols to the molecular alteration of wheat. To include the generation of new wheat germplasm and testing of genes related to wheat functionality, and biotic and abiotic stresses. Improve the efficiency and applicability of wheat transformation and the predictability of transgene expression.
Approach:
1) Use genetic transformation to produce wheat lines with modifications in genes important for end-use properties and agronomic performance, including genes for seed proteins and pathogen resistances. 2) In collaboration, cross transgenes into adapted wheat varieties and evaluate expression stability during field growth and test the effects of transgenes on agronomic performance. 3) Collaborate to test genes for their functionality in transgenic wheat. Candidate genes will be transformed into wheat in order to evaluate the effects of overand under-expression of grain end-use properties and changes in plant resistances to biotic and abiotic stresses. 4) Identify and characterize gene promoter and other DNA regulatory sequences to improve the control of transgene expression. 5) Modify wheat transformation protocols and approaches to increase the efficiency of recovering transgenic wheat lines and to extend transformation to elite germplasm.
