Research Project: Functional Approaches to the Control of Babesiosis in Cattle and Horses

Objective:
1) To determine the genes responsible for pathogenesis and transmission, babesia species will be isolated at various stages of infection to determine expressed genomic sequences.

Approach:
Our plan involves acquisition of the genome sequence of B. bovis and identification of B. bovis and B. equi tick-midgut specific antigens, functional characterization after gene inactivation by transfection techniques, and the teting of their ability to block transmission when administered as vaccine. Definition of these genes and the use of the transfection system will also allow the production of genetically defined transmission-deficient attenuated strains that could be employed as improved vaccination tools. Our selection criteria for a transmission blocking vaccine candidate will be ability to elicit antibodies able to block midgut infection, and for inability of the parasite to replicate in the tick midgut following functional knockout of the specific gene.
