PCP Immunoassay Technologies (1.02MB)

U.S. EPA Superfund Innovative Technology Evaluation (SITE) ProgramEnviroGardTM PCP Immunoassay Test Kit1, Penta RISc Test System2, PCP RaPID Assay2
Millipore Corp1., and Stategic Diagnostics, Inc.2

Abstract

Three enzyme-linked immunosorbent assay technologies for pentachlorophenol (PCP) testing in soil and water were evaluated. Penta RISc Test Systems (formerly ENSYS, Inc.), EnviroGardTM PCP Immunoassay Test Kit (Millipore Corp.), and Pentachlorophenol RaPID Assay (formerly Ohmicron Corp.) are designed to quickly provide semiquantitative results. Antibodies fix to the inside wall of a test tube, where they offer binding sites for PCP. An enzyme conjugate containing a PCP derivative is added to the test tube to compete with sample PCP for antibody binding sites. Excess sample and enzyme conjugate are washed from the test tube. Reagents are then added to the test tube to react with the enzyme conjugate, forming a color. After a designated time, a solution is added to the test tube to stop color formation. The color is compared to the color formed by a PCP standard. All three kits were evaluated at sites in Morrisville, NC and Winona, MO in the summer of 1993. The EnviroGardTM kit did not meet accuracy requirements; the PCP analysis protocol has since been revised.
