Research Project: Transfer of Total Sclerotinia Resistance from Phaseolus Coccineus to P. Vulgaris

Objective:
Develop white mold resistant dry and snap beans using highly resistant accessions of Phaseolus coccineus as a source by transference of complete resistance from P. coccineus to P. vulgaris using a mapped based marker assisted selection strategy.

Approach:
Resistance QTLs will be mapped in a PI25 x WP population. About 150 F2 individuals are available and have been previously characterized for resistance using the straw test. The test will be repeated to verify previous results and identify escapes. DNA will be isolated from young leaves. Primer ID and PCR amplification will be used. Newly identified SSR markers will be evaluated and placed on the map. A second mapping population from OR91G x 25 will be developed to identify possible factors not present in common bean associated with white mold resistance. Knowledge from studies of disease mechanisms will guide the ID of markers.
