Research Project: Resistance Improvement of Bean Through Multi-Site Screening and Pathogen Characterization

Objective:
1) identify sources of resistance to Sclerotinia sclerotiorum in adapted common bean lines, and 2) assess the genetic variation in common bean isolates of S. sclerotiorum.

Approach:
White mold reaction of putative sources of resistance will be determined in field, laboratory and greenhouse screening tests. Screening methods will be correlated over multiple locations and investigators. Mycelial compatibility groups of isolates from the major bean growing areas will be determined. Aggressiveness (similar to virulence) will be determined by inoculation of each isolate onto detached leaves of bean lines with partial resistance. Polymorphisms in ribosomal DNA lengths to discriminate subspecies variation in S. sclerotiorum. Microsatellite-specific primers will characterize the intraspecific variation in S. sclerotiorum field isolates collected at each screening trial site.
