Research Project: Validation and Introgression of a White Mold Resistance Gene from Andean into Middle American Germplasms

Objective:
1) determine the level of expression of the physiological resistance QTL in a different genetic background to verify its utility and to provide resistance in adapted pinto beans; 2) create a more saturated map in the regions flanking the QTL; and 3) determine if linkage between the QTL and the T phaseolin locus can be broken.

Approach:
Use recombinant inbred line (RIL) populations developed at Colorado St. Univ. to validate the QTL and attempt to find new markers tightly linked to the QTL. Characterize a recombinant inbred population from the cross G122 x CO72548. The RIL population will be screened using the straw test to identify resistant lines and the phaseolin marker will be evaluated for linkage with resistance among the lines. Additional random amplified polymorphic DNA , amplified fragment length polymorphism, simple sequence repeats, or inter-simple sequence repeat markers will be identified.
