Research Project: Identification and Development of Resistance to Whitefly-Transmitted Tomato Criniviruses

Location: Crop Improvement and Protection Research

Project Number: 5305-22000-010-02
Project Type: Reimbursable

Start Date: Apr 01, 2000
End Date: Feb 28, 2005

Objective:
Evaluate germplasm for resistance to TICV and ToCV, in order to identify sources of resistance against these and other criniviruses in tomato. Continue development of tomato expressing specific viral genes in efforts to produce engineered resistance against tomato criniviruses. Improve detection methods for tomato criniviruses to allow detection of lower levels of virus than is currently possible. Present methods often require passaging of virus to propagative hosts prior to detection.

Approach:
Wild and segregating tomato lines will be provided by university cooperators. Greenhouse whitefly (Trialeurodes vaporariorum) populations will be increased and used to transmit TICV to young tomato seedlings. Plants will be tested for resistance by PCR and hybridization to virus specific probes. Tomato plants will be transformed with select viral gene sequences to obtain resistance and for exploring virus vector relationships. Documents Reimbursable with CA Tomato Board. Log 17546. BSL-1; 02/2003. Formerly 5305-22000-007-02R (9/03).
