Objective:
Analysis of molecular changes upon exposure of prostate or breast epithelial cells to tomato-derived phytonutrients using R7-PCR and CDNA microarray technology.
Approach:
The nutrients of interest will be purified using high-pressure liquid chromatography (HPLC). Normal and cancerous prostate and mammary cell lines including LnCaP, DU145 and MCF-7 will be maintained in culture systems. The optimal concentration of nutrients for the alternation of biomakers including phase I and phase II enzyme activities will be determined using dose curve with various concentrations of nutrients supplemented in the culture media. USDA has the facilities to measure cytochrome P450 1A1 (CYP1A1) and NAD(P)H: quinone oxidoreductase 1 (NQOR1). Dr. Thomas Wang, Research Chemist of the Phytonutrients Laboratory, will evaluate the molecular effects of the carotinoids of interest using RT-PCR and cDNA microarray technology in model cell culture.
