Objective:
Determine the etiology and epidemiology of Phytophthora root and crown rots, bacterial, virus and virus-like disease of tree fruits and nuts, grapevines and strawberries. Develop improved reliable methods to detect and identify the pathogens associated with these diseases. Develop safe, effective, environmentally sound disease management strategies, including alternatives to methyl bromide pre-plant soil fumigation. Determine factors involved in physiology/biochemistry of resistance and susceptibility to Xf through interaction with ARS scientists at Parlier, California. Refine PCR tests and examine applicability for advanced diagnostics. Develop and field test molecular assays for Sudden Oak Death and develop marker assays. Identify new regions of the P. ramorum genome and initiate bioinformatics with scientists at Fort Detrick, Maryland and Salinas, California.
Approach:
Isolate and identify pathogens associated with important emerging diseases of almond, walnuts, stone fruits, grapevines, and strawberries. Determine the etiological roles of these pathogens in specific diseases. Determine means of pathogen spread and identify biotic and abiotic factors that affect infection and disease development. Adapt and develop improved biological, serological, and molecular techniques for reliable pathogen detection and identification in planting stock and propagating material. Evaluate cultural practices (e.g. sanitation, soil water management), host resistance and available chemicals to control these diseases, especially as alternatives to methyl bromide soil fumigation to control soilborne diseases.
