Objective:
Determine the nucleotide sequence of the genome of Spiroplasma kunkelii, causal agent of corn stunt disease. Identify genes, promoters, regulatory and other genetic elements in the genome. Determine the organization of the genome. Identify metabolism related genes including those forming complete or incomplete metabolic pathways.

Approach:
Carry out whole genome shotgun cloning and sequencing of genomic DNA subclones into double-stranded sequencing vectors in two phases. Prepare large insert Cosmid, BAC, PAC, or other subclones and end sequence clones. Carry out gap closure and finishing, involving custom synthetic primer walking using a combination of double-stranded shotgun subclones and large insert end-sequenced subclones, and PCR-based closure reactions. Carry out detailed annotation of the genome sequence for publication and submission to GenBank. BSL-1 Recertified 9/13/01.
