Objective:
Sequence and annotate the genome of Spiroplasma kunkelii. Identify genes useful for spiroplasma species- and strain-level detection and identification. Expand the molecular scheme for phytoplasma classification. Delineate criteria for distinguishing among phytoplasma species and establishing a formal genus Phytoplasma taxonomy. Identify and characterize functions of pathogenesis genes in spiroplasma and phytoplasma. Develop PCR-based detection and identification of Ralstonia solanacearum, biovar 2/race 3, causing brown rot of potatoes.
Approach:
Gene sequencing, bioinformatic methods for sequence analysis and gene discovery, recombinant DNA techology, phylogenetic and gene evolution analyses, tissue culture, enzyme and bacterial growth assays, gene function analyses, and biochemical methods will be employed with inoculations of susceptible plants, tissue culture and plant transformation, to investigate the structure and function of DNA sequences and gene products as molecular markers for the detection and identification of species- and strain-level evolutionary divergence and as potential targets for design of disease control measures.
