Objective:
The first onbjective is to map between 200 and 250 new microsatellite markers in the wheat genome via recombination mapping in the International Tritici Mapping Initiative (ITMI) mapping population. The second objective is to use a set of selected microsatellite markers to characterize the potent scab resistant genotypes Sumai 3, Ning7840 and W14 along with a panel of elite U.S. breeding parents.
Approach:
DNA of the parents and the 105 recombinant inbred lines (RILs) of the ITMI mapping population will be isolated. Newly developed microsatellite markers that are polymorphic in the ITMI mapping population will be sent to Michigan State Univ. and will be positioned in the wheat genome via recombination mapping in the ITMI RILs. DNA will be isolated from the scab resistanct cultivars Sumai 3, Ning7840 and W14 as well as a panel of 36 hexaploid and durum wheat breeding parents identified by breeding programs in scab-threatened areas of the U.S. Mapped microsatellite loci will be assigned to one of 84 arm sectors (2 per chromosome arm one distal and one proximal to the centromere). Allelic states for the two loci in each of the arm sectors will be determined for each accession using fluorescent labelled nucleotides analyzed on a Perkin-Elmer 377 DNA sequencer.
