Objective:
The overall objective of this research is the creation of a single nucleotide polymorphism (SNP)-based genetic map of the soybean. The specific objective of the research is the genetic mapping of SNP DNA markers and the application of the resulting genetic map for the discovery of genes that are useful for the genetic improvement of soybean.

Approach:
Polymerase chain reaction (PCR) primers will be designed to existing soybean unigene DNA sequence data from GenBank at the National Center for Biotechnology Information. The PCR primers will be tested via the analysis of the resulting PCR products. Finally, the DNA sequence of PCR products from a small group of soybean genotypes will be determined and compared using PolyBayes SNP discovery software. SNP discovery will be followed by the development of single base extension assays that allow the detection of the SNP DNA markers using the Luminex flow cytometer. Each assay will be optimized to increase the efficiency of SNP detection. The materials needed to conduct each single base extension assay will be forwarded to the Univ. of Nebraska, Lincoln along with all pertinent information needed to successfully use the newly developed DNA markers. Each new SNP DNA marker will be genetically mapped in the University of Utah Minsoy x Noir 1 mapping population. The genetic mapping will require the determination of the SNP allele that is present in each of the 80 members of the mapping population using the Luminex flow cytometer. SNP genotype data will be used to genetically map each SNP DNA marker on the soybean genetic map.
