Objective:
Cooperative research will determine: appropriate germplasm sample size; vulnerability of genes to accidental loss; association of RAPD markers with useful traits; uniformity of genetic diversity assessment using different tissues; inadvertent selection in routine protocols; and parity of presumed duplicates among world genebanks.
Approach:
Model Solanum species populations from the US Potato Genebank will be grown DNA harvested and characterized with RAPD markers: 1&2)frequencies of markers will be determined and theoretical risk of accidental loss of genes will be calculated, 3)bands will be associated with segregation for tuber calcium, 4)banding patterns from different tissue sources will be compared, 5)banding of first and last seedlings to germinate will be compared, 6) presumed duplicate seedlots from Russian and US potato genebanks will be obtained and compared.
