Objective:
Determine the role of the nonstructural P17 and structural RT luteovirus proteins in the systemic infection of host plants, and determine the biologically active regions of the luteovirus capsid and RT proteins that regulate the uptake and transmission of luteoviruses by aphids.
Approach:
Three virus genes translated from a common subgenomic RNA are conserved among all luteoviruses and encode multifunctional proteins that are involved in virion structure, aphid transmission, and movement within plant hosts. A functional genomics approach will be used to determine which protein sequences are required for each known function of the protein. Using an infectious clone of potato leafroll virus and agrobacteria-mediated infection of plants, methods were developed that will allow us to rapidly screen genetically altered viruses for their ability to assemble stable virions, to systemically infect plants, and to circulate in and be transmitted by aphid vectors.
