Research Project: Addressing the Pvy Challenge in the Western Us: a Practical Approach

Objective:
Development of definitive molecular methods to characterize PVY virus strains. Deployment of reliable and inexpensive diagnostic procedures that can be used by seed certification agencies to detect and distinguish PVYn from PVYo in multiplex assays. Conduct of a comprehensive survey of potato seed lots, commerical fields, and potential weed hosts to determine the incidence of PVY strains and determine the symptomatology and effects of infection on our most important varieties.

Approach:
Characterize the biology and molecular characteristics of PVY strains that have been found in the US, including conclusive genotyping of all distinct strains. Develop a genetic lineage tree to help determine if sources of variation are from introductions or natural virus recombination. Use bioassay on potato, tobacco, and other hosts to support RNA sequence data. This will include a complete review of information and characterization of the purported PVYntn strain found in California.
