Objective:
1) Determine the expression profile of SCN Hsp90 mRNA and protein during nematode development. 2) Determine the protein partners that interact with SCN Hsp90 to aid in its function during development. 3) Determine whether natural products or microbial inhibitors of Hsp90 interfere with SCN develoment, and determine whether these methods can be applied to effectively control SCN.

Approach:
1) Produce the Hsp90 protein in bacteria, make antibodies to the protein. Use the anti-Hsp90 antibodies to track Hsp90 levels during the development of SCN from egg to adult, compare Hsp90 levels in different SCN strains or populations, and track changes in Hsp90 when nematodes are exposed to heat, inhibitors, or other stressors. 2) Test Hsp90 inhibitor geldanamycin for effects on SCN hatching, juvenile mobility, or other traits. 3) Test the interaction of the SCN hatching proteins within the HCH-1 complex with the Hsp90 protein, using a rapid yeast two-hybrid sceening system. 4) Use the yeast two-hybrid screening system to test the interaction of SCN Hsp90 with developmental arrest ("DAF") gene products from the non-parasitic nematode C. elegans, in order to determine the function of Hsp90 in SCN and how it may differ from the non-parasite form.
