Objective:
1. Improve and modernize the classification of plant-parasitic nematodes and introduce new taxonomic techniques and characters into nematode systematics; 2. Provide expert nematode identifications and curatorial services involving the USDA Nematode Collection, as urgently required by ARS scientists, federal and state researchers, and regulatory agencies.

Approach:
1. Determine new characters, including molecular sequences of the heat shock chaperone gene hsp90 and different regions of ribosomal genes, and their use with species-specific primers or real-time PCR probes where appropriate. Associate thermal recovery assays and physical density differences with morphological and molecular characterizations and apply both to different degrees among various populations and species of nematodes of economic and regulatory importance. 2. Streamline the use of traditional morphological characters for identification and classification by new applications to existing populations and new plant interceptions.
