Objective:
Develop a single assay that can detect and differentiate multiple bacterial potato pathogens and that has the capacity to detect emerging pathogens for which no sequence information is available.

Approach:
Complete proof-of-concept experiment with Erwinia, Pantoea, and Pseudomonas by sequencing the PCR products to confirm that the target sequences have been amplified; determine, using SSCP, if species in these genera can be distinguished; develop a preliminary database based upon the DNA fragment migration profiles; and; determine if this method can detect and differentiate Erwinia in soft rot samples collected and frozen during the potato harvests of 2002 and 2003.
