Objective:
Determine the allelism of four putative novel sources of root-knot nematode resistance in grape with the N resistance allele.
Approach:
The relationship of four putative novel sources of root-knot nematode (Meloidogyne) resistance to the N allele will be assessed. Genetic analysis of root-knot nematode resistance in grape will be completed using greenhouse screening to determine the nematode resistance of two generations of grapevine seedlings, followed by the development of a model of genetic control consistent with the observations. Sources of the four putative novel alleles will be crossed to grape accessions homozygous for the N allele. Nematode resistance of first generation seedlings will be evaluated in pot culture in a greenhouse, with nematode reproduction measured by counting eosin-Y stained egg masses. Double heterozygote seedlings will be forced to flower in the greenhouse and selfed or sibling mated. Segregation patterns shown by nematode resistance screening of second generation seedlings will reflect allelic relationships between the four putative novel sources and the N allele.
