Objective:
To develop and test protocols for in situ hybridization histology of Xylella fastidiosa in infected grape tissues.

Approach:
Two established methods will be modified and blended to achieve DNA-DNA in situ hybridization of Xylella fastidiosa bacteria in sectioned grape tissue, for epifluorescent and confocal visualization of bacteria in planta. RNA-RNA hybridization of sectioned plant material, and DNA-DNA hybridization of unsectioned, cleared and macerated cells will be used. Other, literature-based methods will also be consulted and portions added as needed or appropriate. Documents SCA with U. of MO.
