Research Project: Genetic Manipulation of Destruxins in Metarhizium Anisopliae

Location: Plant Protection Research Unit

Project Number: 1907-22410-003-06
Project Type: Specific C/A

Start Date: Jan 01, 2003
End Date: Dec 31, 2004

Objective:
Use genetic manipulation to generate mutants unable to synthesize destruxins. Clone and sequence destruxin synthetase. Analyze effects of genetically-altered destruxin production on fungal development.

Approach:
To use RT-PCR to confirm that a specific gene is expressed concurrently with destruxin production, to create transformants in which the destruxin peptide synthetase gene has been insertionally inactivated by direct gene replacement. Clone and sequence the complete gene. Determine whether genetically modified transformants are altered in fungal morphology and development.
