Research Project: Enhanced Resistance to Scab by Genetic Engineering with Pr-Protein Genes

Objective:
The objective for this specific cooperative agreement is to facilitate research on development of scab resistant wheat germplasm by development and evaluation of transgenic wheat lines overexpressing different combinations of PR-proteins.

Approach:
Bioassays of currently available transgenic plants for scab resistance will be carried out using the single floret inoculation assay in the greenhouse as well in the scab field nursery using well established protocols. New transgenic plants containing other PR-protein genes will be generated using a biolistic approach using the bar selection. Crossing of the transgenic plants will be carried out and progeny will be analyzed for the presence of all transgenes and their expression will be followed using western blotting. Progeny containing the desired combination of transgenes will be tested for resistance to scab.
