Summary:
Large samples (> 1L) are usually required and concentration by the membrane filter technique (SM-9260B section 1.d) is recommended although other methods are also acceptable. Samples are proportioned into a five-tube, three-dilution set using either selinite cystine, selenite-F, or tetrathionate broth as the selective enrichment medium (SM-9260B section 3). After incubation as necessary, plates from each tube are streaked on plates of brilliant green and xylose lysine desoxychlolate agars followed by 24 h incubation at 35 deg. C. Several colonies suspected of being Salmonella are selected from each plate and a slant each of triple sugar iron and lysine iron agars are inoculated and then incubated for 24 h at 35 deg. C. Test those cultures giving a positive reaction by serological techniques (SM-9260B section 5). The MPN/1.0 L of original sample is calculated from the combination of Salmonella negative and positive tubes. Concentration by the membrane filter technique (SM-9260B section 1.d) is recommended although other methods are also acceptable. Incubation Time: 72 h. Technique: Culture.
